The essential requirement for anaerobic digestion of industrial wastes is that the process should operate reliably at high performance. In the digestion of dilute, soluble wastes it is necessary to retain the active biomass within the digester at short liquid retention times for the process to be economically feasible and this is reflected in digester design. Performance of digesters can only be assessed by interpretation of measurable parameters such as pH2, Eh, pH, volatile fatty acid concentrations, temperature, gas production, biomass content and feed rate and composition. The effects of changes in these parameters on the microbiology of methanogenic digestion and the application of this knowledge in control of the process is discussed.  相似文献

AIMS: To combine database-held sequence information with a programme of experimental molecular ecology to define the methanogenic community of a hypereutrophic lake by a PCR-restriction fragment length polymorphism (RFLP) analysis. METHODS AND RESULTS: Methanogen diversity in a hypereutrophic freshwater lake was analysed using 16S rDNA PCR-RFLP. Database-held 16S rRNA gene sequences for 76 diverse methanogens were analysed for specific restriction sites that permitted unequivocal differentiation of methanogens. Restriction digestion and agarose gel electrophoresis of the 16S rDNA from selected methanogen pure cultures generated observed restriction profiles that corroborated the expected patterns. This method was then tested by analysing methanogen diversity in samples obtained over 1 year from sediment and water samples taken from the same sampling site. CONCLUSIONS: Restriction analysis of the 16S rRNA gene sequences from 157 methanogen clones generated from lakewater and sediment samples showed that over 50% were similar to Methanoculleus spp. Furthermore, a total of 16 RFLP types (1-16) were identified, eight of which contained no cultured representative archaeal 16S rRNA gene sequences. SIGNIFICANCE AND IMPACT OF THE STUDY: This RFLP strategy provides a robust and reliable means to rapidly identify methanogens in the environment.  相似文献

全球变暖是全人类面临的一个巨大挑战,而温室气体排放持续上升是全球变暖的关键因素,并引发一系列生态环境问题。甲烷是第二温室气体,对全球变暖的贡献达20%。然而,在甲烷代谢中发挥重要作用的产甲烷古菌和厌氧甲烷氧化古菌(anaerobic methanotroph,ANME)较难培养,极大地限制了人们对甲烷代谢及其影响碳源-汇关系与机制的研究。本文综述了最新产甲烷古菌和ANME富集、分离和培养方法,包括富集培养、原位培养、共培养、微流控技术、稀释分离和固体分离技术、ANME反应器和培养瓶富集培养,以及宏基因组预测和反向基因组学,并对这些方法的优缺点进行了评估,对未来甲烷代谢古菌的富集、分离和培养提出新的建议。  相似文献

Archaea are renowned for their ability to thrive in extreme environments, although they can be found in virtually all habitats. Their adaptive success is linked to their unique cell envelopes that are extremely resistant to chemical and thermal denaturation and that resist proteolysis by common proteases. Here we employ amyloid-specific conformation antibodies and biophysical techniques to show that the extracellular cell wall sheaths encasing the methanogenic archaea Methanosaeta thermophila PT are functional amyloids. Depolymerization of sheaths and subsequent MS/MS analyses revealed that the sheaths are composed of a single major sheath protein (MspA). The amyloidogenic nature of MspA was confirmed by in vitro amyloid formation of recombinant MspA under a wide range of environmental conditions. This is the first report of a functional amyloid from the archaeal domain of life. The amyloid nature explains the extreme resistance of the sheath, the elastic properties that allow diffusible substrates to penetrate through expandable hoop boundaries, and how the sheaths are able to split and elongate outside the cell. The archaeal sheath amyloids do not share homology with any of the currently known functional amyloids and clearly represent a new function of the amyloid protein fold.  相似文献

冻土是陆地生态系统中最容易受到全球气候变化影响的碳库,既发挥着碳源又起着碳汇的作用。人们非常关注贮存于冻土中有机碳的最终归宿,是因为全球气候变暖会加快冻土的解冻,释放更多的温室气体(二氧化碳和甲烷)到大气中,从而进一步加剧温室效应。据估计每年从北半球冻原陆地生态系统释放进入大气的甲烷约占全球自然界释放甲烷总量的25%。研究证实冻土生物源甲烷的产生和消耗分别由耐(嗜)低温的产甲烷菌(methanogens)和甲烷氧化菌(methanotrophs)介导。鉴于冻土甲烷循环对全球甲烷平衡的显著作用以及在冻土生物地球化学循环中的重要功能,对介导冻土甲烷循环的产甲烷菌和甲烷氧化菌的研究将有助于更好地评估冻土生态系统对全球气候变化的响应和影响,本文就冻土甲烷循环过程、产甲烷菌、甲烷氧化菌的群落结构、活动、生态功能及其对气候和环境变化的响应机制的最新研究进行综述,以期为我国开展冻土甲烷循环机理研究提供支持。  相似文献

A quantitative fluorogenic PCR method for detecting methanogenic and methanotrophic orders was established using a refined primer set for the methyl coenzyme M reductase subunit A gene (mcrA). The method developed was applied to several microbial communities in which diversity and abundance of methanogens or anaerobic methanotrophs (ANMEs) was identified by 16S rRNA gene clone analysis, and strong correlations between the copy numbers of mcrA with those of archaeal 16S rRNA genes in the communities were observed. The assay can be applied to detecting and assessing the abundance of methanogens and/or ANMEs in anoxic environments that could not be detected by 16S rRNA gene sequence analyses.  相似文献

The lysine requirements of rats of various body weights were estimated using the feeding and isotope tests.

The regression equation obtained by the feeding test was Y= 1.03 – 0.58 log X. Where Y is lysine percentage of the diet and X is the mean of initial and final body weights (g) of rats achieving optimal growth gains during the feeding period.

The regression equation obtained by the isotope test was 7=0.90 – 0.49 log X, where Y and X are lysine percentage in the diet and body weights (g) of rats achieving optimal growth gains at the injection time respectively.  相似文献

Various anaerobic hydrolytic and methanogenic bacteria active in cattle dung biogas plants are reported in the literature. Anaerobic bacteria with ability to use volatile fatty acids constitute a vital bridge between hydrolytic bacteria and methanogenic bacteria. The present paper describes the isolation ofSyntrophobacter wolinii a propionate degrading bacterium in co-culture with a hydrogen utilizing methanogenviz.,Methanobacterium formicicum from the fermenting slurry of cattle dung biogas plant. Earlier studies on propionate and butyrate degradation indicatedMethanospirillum hungatei as the hydrogen utilizing partner of the co-culture whereas in the present studies this was not the case. Temperature 35° C, pH 7.5 and 20 mM of propionate were found optimal for growth and activity of co-culture.  相似文献

Metal cofactors are required for many enzymes in anaerobic microbial respiration. This study examined iron, cobalt, nickel, copper, and zinc in cellular and abiotic phases at the single-cell scale for a sulfate-reducing bacterium (Desulfococcus multivorans) and a methanogenic archaeon (Methanosarcina acetivorans) using synchrotron X-ray fluorescence microscopy. Relative abundances of cellular metals were also measured by inductively coupled plasma mass spectrometry. For both species, zinc and iron were consistently the most abundant cellular metals. M. acetivorans contained higher nickel and cobalt content than D. multivorans, likely due to elevated metal requirements for methylotrophic methanogenesis. Cocultures contained spheroid zinc sulfides and cobalt/copper sulfides.  相似文献
